Suggestions for the new room.
Double studs, staggered, so you can soundproof the place with non-lining-up vibration nodes. Stick a sheet of drywall between the two 2X4 walls that is screwed to alternate sides.
Fill the spaces with high-density fiberglass or rock wool insulation that is acoustic and thermal.
Double ceiling, first layer of drywall screwed securely to the ceiling, second layer to S-bars running across the ceiling rafter pattern. Ceiling also well-insulated.
Floor: nice, sturdy hardy-backer with stone or ceramic tiles on it.
Steel door with foam core. Plenty soundproof.
Outlets oriented sideways instead of vertically, every couple of feet along the wall, integrated into a tall baseboard. Receptacles that are isolated circuits, with isolated neutrals, 20 amps, not 15. Lights on 15 amp circuit, separate. If you really like electrical quality, run wires in conduit, and make the ground wire and neutral double the ampacity of the feeders for the circuits.
Just some construction ideas. Nice to be able to game away with aplomb, without disturbing anyone in the house.
Have fun with all that Morto.
Good suggestions as well Traak. When I ever get to building a place I do plan on doing something akin to that.

Thanks, Goku. Oh, more details: never have one stud space with a penetration, such as an electrical box, that backs into another stud space with a penetration in the next room, because then people can hear right through one electrical box into your gaming sanctum.
A vestibule with about four feet of space leading to a second door will also minimize unwanted sound exfiltration.
